3. CONTENT DESCRIPTION

THEORY

PART I: Manipulation of nucleic acids in vitro. Basic techniques.

1. Introduction to genetic engineering. Basic techniques to manipulate nucleic acids in vitro. Historical perspective of the technology of analysis and manipulation of nucleic acids. Purification of DNA and RNA. Separation and analysis of nucleic acids by gel electrophoresis.

2. Enzymes used to manipulate nucleic acids in vitro. Cut and joining of DNA. Chemical modification in vitro. Synthesis and degradation of NNAA.

3. Amplification of NNAA in vitro (PCR) and applications. Polymerase Chain Reaction (PCR): Taq polymerase, primers and amplification. PCR using RNA (RT-PCR). Applications of the PCR.

4. Nucleic acid hybridization applied to sequence identification. Transfer of NNAA to membranes. Probes. Hybridization. Detection.

5. Gene expression analysis. Northern-blot, RNase protection assays, RT-PCR, in situ hybridization, primer extension assay.

PART II: Genetic manipulation in prokaryotes ( E. coli)

6. Cloning and expression in prokaryotes ( E. coli), Part I: Introduction to cloning. Cloning vectors commonly used in prokaryotes. Methods to introduce new genetic information in prokaryotes.

7. Cloning and expression in prokaryotes ( E. coli), Part II: Detection and identification of positive clones: selection of recombinants.

PART III: Genetic manipulation in eukaryotes

10. Genetic manipulation and genetic improvement in microorganisms. Applications. Yeast as eukaryotic model organism. Biotechnological importance of yeast. Genetic engeniering in yeast and other fungi. Examples and applications.

11. Genetic manipulation and genetic improvement in plant. Applications. Classical plant improvement. Genetic manipulation techniques in plants. Transgenic plants. Examples and applications.

12. Genetic manipulation and genetic improvement in animals. Applications. Classical animal improvement. Genetic modification of cells and complete organisms. Growth and production. Some applications.

13. Legislation about the use of genetic modified organisms and patents. Economical, ethical and cultural aspects. Legislation.

 

PRACTICAL LESSONS

A) Laboratory

Laboratory practical lessons will be taught in 2 weeks (2 hour session per day). The procedures performed include several techniques on NNAA manipulation reviewed in theory.

The practices proposed for the next course could change depending on the applicable face-to-face scenario according to the current health situation.

PRACTICE 1 . Southern-Blot. This practical includes learning of nucleic acids labelling techniques, labelling quantification, nucleic acid transfer to nylon membranes, in situ hybridization with a labelled probe and detecting the label by immunological reactions. The objective is to identify DNA specific sequences in genomic or plasmidic samples.

PRÁCTICA 2 . Cloning in E. coli. This practical lesson includes: DNA restriction and ligation (synthesis of recombinant DNA), obtaining bacterial competent cells, transformation of competent cells with recombinant DNA (previously obtained in class) selection of recombinant bacteria, extraction of plasmid DNA, restriction and electroforesis of plasmid DNA and screening for the presence of insert.

Applications of recombinant proteins.

4. COURSE DESCRIPTION AND TEACHING METHODOLOGY

Large group: 3 ECTS

Master classes: Theory classes where a global and integrated vision of the units within each block is provided. At the end of the thematic blocks, the students will work on the level tests: resolution and discussion (when applicable) of short questions about the block.

Small group: 2.8 ECTS

Training activities (0.8 ECTS): Problem solving and case studies, whose resolution requires the assimilation of basic concepts related to the proposed topics. These practical assumptions will be based on real scientific work so that the student will have to reproduce the attitude of the scientist when faced with a specific question as to what methodology to apply to answer it or what the results obtained mean.

Practical (2 ECTS): Practical laboratory sessions with small groups - In these sessions students will carry out different experiments in the laboratory related to the concepts explained in master classes. In the practical lessonsa detailed protocol is provided, where the basis of the practice is explained, together with the process that the student must follow on his own to obtain the desired objective.

Individual/group tutorials: 0.2 ECTS

5. ASSESSMENT METHODOLOGY

Ordinary Call I:

The evaluation will be continuous, and will attend to several levels and different aspects. For this purpose, the score obtained in the different activities developed throughout the course will be considered (the weighting on the final score is indicated in brackets):

- The qualification of the level tests (35%). It will be necessary to reach a minimum of 4 out of 10 in the final total of all the tests in order to pass the course through continuous assessment.

- The grade of the exam (35%). It will be necessary to reach a minimum of 4 out of 10 in the exam to be able to pass the course through continuous assessment.

- The laboratory practices (15%), which will be evaluated according to attendance, understanding and results obtained.

- Training activities (15%): the participation of the student, the dialectic quality that the student shows in his interventions, either through his own questions, or through the contributions to questions raised by the teacher or by his own colleagues, as well as the quality of the work done, will be valued.

The knowledge acquired by the students and their ability to apply this knowledge to specific practical situations will be valued throughout the course with the exam, the level tests, formative activities and laboratory practices.

The exam will include free answer and "test" type questions. The first ones allow to evaluate the capacity of synthesis and relationship, as they force the student to organize and structure his knowledge in a logical way. The latter will evaluate the student's instantaneous response capacity. In this way, the following competencies will be evaluated: CT1, CT3, CT5, CT6, CT7, CT8, CE37, CE39, CE43, CG11, CG8.
On the other hand, with the level tests, the laboratory practices and the training activities will be evaluated
also the competences: CT1, CT2, CT3, CT4, CT5, CT6, CT7, CT8, CT9, CT10, CE37, CE39, CE43, CG12.

10 Practical sessions in practice laboratories

The Faculty initially establishes attendance at 50%, although this will be adapted to allow for maximum attendance with the limitations on laboratory capacity determined in each case by the Occupational Risk Prevention Service.

The number and duration of face-to-face sessions will be established once the effective capacity of the laboratories has been determined by the Occupational Risk Prevention Service and the number of students registered.

2-hour sessions in the laboratory. For those sessions that have to be online, synchronous format will be used in combination with asynchronous activities using the tools provided by the Virtual Teaching and G Suite services.

 

4 Training activity sessions

The Faculty initially establishes 100% attendance, although this may be modified taking into account the number of students enrolled and the limitations on classroom capacity determined by the Occupational Risk Prevention Service.
